TELS1 reveals a minor contribution for telomere loops in chromosome end-protection

Chromosome-end protection is crucial for genome stability, with telomere loops (t-loops) thought to play an important protective role. In most cell types, t-loop stabilization relies on the shelterin protein TRF2, while the regulatory pathways in pluripotent cells remain undefined. We have identified Telomere Loop Stabilizing Protein 1 (TELS1), which stabilizes t-loops independently of TRF2 in pluripotent cells. I will describe how TELS1 likely makes the t-loop in these cells, and I will demonstrate that making a t-loop is insufficient to protect chromosome ends. I will discuss the implications of our findings for telomere biology and genome stability.
